1 Kilobaser preparation

1.1 Unpacking and inspection

Prior to opening the shipping container, inspect it for damage or evidence of mishandling. If it has been damaged or mishandled, notify the carrier before opening the container. Carefully unpack the Kilobaser and accessories and inspect them for completeness and possible damages from transportation. Any damage should be reported to both carrier and supplier immediately. Save the shipping container. Check the contents against the packing list.

em CAUTION
Keep in mind the weight of the Kilobaser device (21 kg; 46 lbs). Be careful when you lift it out of the packaging and carry it; if necessary, ask a colleague for help.

1.2 Scope of delivery

1.3 Location and environment

Place the Kilobaser one or one-XT on a stable, flat and dry surface. There should be at least 15 cm (6 in) clearance between the wall and the rear of the Kilobaser to guarantee sufficient ventilation and to assure enough space to connect or disconnect electrical and gas connections. The clearance above the device must not be less than 25 cm (10 in) to allow for unconstrained opening of the lid. The acceptable environment for Kilobaser is normal indoor laboratory conditions. The area must be clean and well-ventilated; preferable location is a fume hood, where good suction can remove gases that are released during the final drying step. Temperature should be stable and humidity should not be too high, as DNA synthesis is moisture-sensitive. The specific temperature and humidity conditions are 15 to 40 °C and 0% to 60% relative humidity.

1.4 Electrical connections

Connect the Kilobaser one or one-XT to a 100-230V AC, 50-60 Hz power socket using the supplied external power supply unit. Input voltage is 24 V/8 A. External power supply is allowed to be connected to mains supply with circuit breaker 16 A or circuit breaker 20 A in USA/Canada.

Use only the power supply included in the delivery! In case of damage, contact Kilobaser support to provide you with a new one.

Take care to connect the power supply to the Kilobaser device before plugging the power supply into the mains socket!

1.5 External touchscreen display

Kilobaser is equipped with a separate, movable touchscreen display. It allows for easy access to local operations without login credentials, as well as configuration of remote connection and integration into your network via Ethernet or WiFi. Connect the display with the supplied HDMI and USB cables by plugging them into the respective rear ports of the Kilobaser. Make sure to connect the USB-C cable into the lower port on the display. If you connect the cable into the upper port of the display (charging port), the touch function of the display will not work.

1.6 Switching on/off

To switch the device on, press the ON/OFF button. The LED on the button lights up. After a couple of seconds of booting up, the connected touchscreen display shows the user interface.
To switch the device off, press the ON/OFF button and wait until the blinking of the LED stops. Switch the Kilobser off only when the placeholder is inserted.

em CAUTION
Do not switch off the device while in operation. Doing so will lead to unsuccessful synthesis and in worst cases cause contamination of the reagent cartridge.

2 Gas supply

For Kilobaser to function properly, a gas source with a pressure between 500 kPa and 1000 kPa (5 to 10 bar or 73 to 145 psi) must be connected to the push-in connector on the Kilobaser, using a 6 mm O.D. flexible pneumatic tubing. To connect the Kilobaser to a gas supply, the following is required:

  • Gas source (not supplied by Kilobaser)
  • Pressure regulator with pneumatic fitting
  • Pneumatic tubing

2.1 Gas source

While in principle any inert gas is suitable, it is recommended to use dry argon or nitrogen. Connecting less pure gas or air may result in higher moisture in the system and consequently decrease purity of the synthesized DNA. Many laboratories receive inert gas from a central gas supply through pipelines, while some have a safety cabinet where pressurized gas cylinders are kept.

em CAUTION
If there is no central gas supply and no safety cabinet in the laboratory, keep in mind that compressed gas cylinders must be stored upright and according to hazard classification in a well-ventilated, above-grade, weather-proof storage area that is situated at a safe distance from combustible materials, ignition sources, or intense heat.

Gas cylinders can be obtained in several different sizes. For an idea: a 50 L, 10 L or 1 L compressed gas cylinder (all 200 bar) will suffice for approximately 1000, 200 or 20 syntheses, respectively. While gas cylinders can be bought from numerous suppliers, we recommend a dealer that offers services such as refill, exchange and pickup of empty gas cylinders.

2.2 Pressure regulator

In case of a central gas supply, outlet points are usually fitted at the end of the pipeline to set pressure. Safety cabinets are normally equipped with pressure control panels. In a situation where a pressure regulator is not available in your laboratory, use the regulator included in the delivery.

Please note that the included pressure regulator only fits the argon gas cylinders! If you plan to perform syntheses with nitrogen, use a regulator that fits nitrogen gas cylinders or contact Kilobaser for support.

em CAUTION
Setting an appropriate pressure is very important. It has to be set between 500 and 1000 kPa (5 and 10 bar ; 73 and 145 psi). A lower pressure will cause Kilobaser to stop preventively to protect the cartridge. Since standard pneumatic tubing and fittings are usually pressure resistant only up to 10 bar, a higher pressure may result in gas leakage.

2.3 Pneumatic fittings

The type of outlet connection may differ significantly whether it is the outlet of a pipeline, a pressure regulator or a fume hood gas fitting. Kilobaser has a push-in connector to fit a 6 mm O.D. pneumatic tubing, so it is important that the 6 mm tubing can be connected to the outlet connection of the source. If the tubing cannot be fitted directly, a suitable pneumatic fitting or connector is needed. One side of the pneumatic connection of the fitting has to be a push-in type for a 6 mm O.D. pneumatic tubing, while the other side has to fit onto the outlet connection of the source. The outlet of a pressure regulator is normally equipped with a nozzle with a thread, which can be screwed off and exchanged for a suitable pneumatic fitting (one side the appropriate thread that fits the pressure regulator outlet, and the other side a push-in type for a 6 mm O.D. pneumatic tubing).

The included pressure regulator already has the appropriate fitting installed.

2.4 Pneumatic tubing

The gas source and Kilobaser are connected with a 6 mm O.D. flexible pneumatic tubing.

3 m (10 ft) of pneumatic tubing is included in the delivery.

3 Remote web interface

If you do not want to use the touchscreen or have several devices in close proximity, Kilobaser can be completely remote-controlled from a PC or a smart device through its web interface. To set up the remote-control, please use the touchscreen display for initial preparation.

First, connect the Kilobaser one or Kilobaser one-XT to your network using either a WiFi or Ethernet connection. For a WiFi connection, first make sure that the supplied WiFi USB dongle is plugged into one of the back USB ports and then connect to your WiFi via the page found in the settings/network. For an Ethernet connection, connect the device's rear square RJ45 port to one of your RJ45 network outlet ports or router with an Ethernet cable (not supplied).

Next, the visibility of the device has to be enabled in the settings/security. Then, edit an existing user's login name and password or create a new user in the settings/users.

Finally, in the browser of your PC or smart device, enter the Kilobaser's network address (e.g. https://IP_ADDRESS or https://YOUR_HOSTNAME.lan). The IP address and the hostname of the device are found in the settings/network.

When you connect remotely to your Kilobaser for the first time, a security warning screen should be shown. The warning screen is caused by the self-signed certificate, since internal network-names cannot be certified by external certificate-authorities. The TLS certificate is however necessary to make a secure HTTPS connection to your Kilobaser, encrypting your data between your device and Kilobaser. You may ask your network administrator to resolve the issue by installing a locally-signed certificate. Alternatively, to continue, press “Advanced...” to access additional options. Now click on “Accept the Risk and Continue” (Firefox) or “Proceed to (YOUR HOSTNAME) (unsafe)” (Chromium/Chrome). You will be greeted by the Kilobaser login screen. Log in with the login name and password of the created user. We still recommend that you inform your network administrator, in case your company's network policy enforces localy signed certificates.
