1 Tools and Methods

1.1 Required Tools

These tools are not included in the kit.

• Magnetic rack
• 1x 0.5 - 1 mL tube
• 150 µL dH2O (nuclease free) or 150 µL TE buffer
• Vortex
• Spin centrifuge

1.2 Magnetic Beads System

This purification kit is based on the precipitation of DNA on magnetic beads. These beads can be immobilized in a magnetic field generated by a magnetic rack. Thus, the liquid phase can be easily removed without losing beads. Since only oligonucleotides are precipitated and immobi- lized on the magnetic beads, all other components in the sample remain unbound. During the washing step these components are removed. Bound oligonucleotides are eluted with water or TE buffer from the magnetic beads resulting in purified and dissolved oligonucleotides.
